Abstract: In this mini-review we present the concept and the advancement of a novel strategy termed ''ordered single molecule sequencing based on nanomanipulation (OsmSN)'' for whole-genome sequencing. Techniques have been developed to integrate the processes including dissection and isolation of DNA molecules with sufficient spatial resolution together with the subsequent single molecule PCR to achieve the analysis of a particular DNA fragment from a genome DNA molecule. A nanoparticle-assisted PCR protocol was also developed to enhance the specificity and yield of PCR to meet the special requirement of the OsmSN.
