Abstract: Rotaviral gastroenteritis is a significant public health concern, causing an estimated 145,000 deaths annually in India alone. To develop an effective vaccine, a study was conducted using in-silico and reverse vaccinology approaches. Potent epitopes from Indian neonatal rotaviral strains were identified using IEDB server, and studied for properties such as antigenicity, non-allergenicity, non-toxicity, and interferon-γ inducing abilities. The epitopes were also screened for physiochemical properties using the ProtParam tool. A multi-epitope vaccine was constructed docked with Toll-like Receptor 5 molecule (TLR5) by using ClusPro 2.0 server and was further validated by using in-silico tools. Codon optimisation and in-silico cloning were performed using J-Cat server and SnapGene tool, and finally immune simulation was carried out using C-IMMSIM server.
